RESUMO

Objective:To explore the distinctions between different educational levels: stability of learning state, views on and demand for online courses, and further investigate the influencing factors.Methods:A self-designed questionnaire with good reliability and validity was conducted, and all the students in a military medical university were selected by cluster sampling method, and the data were collected by issuing a network questionnaire. SPSS 26.0 was used for data processing.Results:Undergraduates were different from graduates and doctoral students in stability of learning state, the period of home study: doctoral students (2.41±1.30) h < graduates (2.10±1.17) h and undergraduates (2.15±0.99) h; the frequency of using online courses: undergraduates (4.18±1.10) times > graduates (3.29±1.16) times and doctoral students (3.29±0.98) times. In terms of views on online courses, necessity: undergraduates (4.14± 1.01) > graduates (3.93±1.05) and doctoral students (3.78±1.03); familiarity: undergraduates (3.42 ± 0.91) > graduates (3.27±0.97); adaptability: undergraduates (3.79±0.91) > graduates (3.58±0.94) and doctoral students (3.63±0.97); likability: undergraduates (3.36±1.04) > undergraduates (3.25±0.96) and doctoral students (3.17 ± 1.01); teaching effect: undergraduates (2.80±1.04) > graduates (2.67±1.01) and doctoral students (2.61±1.03). In terms of demand for online courses, ideal number: undergraduates (2.52±1.27) > graduates (2.11±1.21) and doctoral students (2.01±1.25); class style: undergraduates (1.77±0.94) > graduates (2.00±0.92) and doctoral students (2.04±1.83). There were statistically significant differences between undergraduates and postgraduates in all dimensions ( P<0.01 or P<0.05). Conclusion:Undergraduates may be more susceptible to academic environment than graduates and doctoral students, showing that the learning stability of undergraduates is the relatively the worst. Undergraduates most recognize online courses, and different educational levels have different demands for online courses.

RESUMO

<p><b>OBJECTIVE</b>To investigate the effect of the fibronectin extra domain A on the aggressiveness of salivary adenoid cystic carcinoma (SACC) cells, via the clustered regularly interspaced short palindromic repeats (CRISPR)/ associated proteins (Cas) system.</p><p><b>METHODS</b>One sgRNA was designed to target the upstream of the genome sequences of extra domain A(EDA) exon and the downstream. Then the sgRNA was linked into plasmid PX-330 and transfected into SACC-83 cells. PCR and DNA sequence were used to testify the knockout cells, and the monoclones of EDA absent SACC cells were selected (A+C-2, A+C-6, B+C-10). CCK-8 cell proliferation and invasion was then tested in control group and the experimental group.</p><p><b>RESULTS</b>The sgRNA was successfully linked into PX-330 plasmid. Part of adenoid cystic carcinoma cells' SACC-83 genomic EDA exon was knocked out, and the knockdown efficiency was above 70%, but the total amount of fibronectin did not change significantly. Three monoclones of EDA absent SACC- 83 cells were successfully selected with diminished migration and proliferation.</p><p><b>CONCLUSIONS</b>The CRISPR/Cas9 system was a simplified system with relatively high knockout efficiency and EDA knockout could inhibiting SACC cell's mobility and invasiveness.</p>

BACKGROUND: Secondary lymphedema is a lifetime risk for breast cancer survivors and can severely affect quality of life. Early detection and treatment are crucial for successful lymphedema management. Limb volume measurements can be utilized not only to diagnose lymphedema but also to track progression of limb volume changes before lymphedema, which has the potential to provide insight into the development of this condition. OBJECTIVES: This study aims to identify commonly occurring patterns in limb volume changes in breast cancer survivors before the development of lymphedema and to determine if there were differences in these patterns between certain patient subgroups. Furthermore, pattern differences were studied between patients who developed lymphedema quickly and those whose onset was delayed. METHODS: A temporal data mining technique was used to identify and compare common patterns in limb volume measurements in patient subgroups of study participants (n = 232). Patterns were filtered initially by support and confidence values, and then t tests were used to determine statistical significance of the remaining patterns. RESULTS: Higher body mass index and the presence of postoperative swelling are supported as risk factors for lymphedema. In addition, a difference in trajectory to the lymphedema state was observed. DISCUSSION: The results have potential to guide clinical guidelines for assessment of latent and early-onset lymphedema.

Objective To establish the quality standard for Fufang Xiongdan Yinchen Granule. Methods TLC was employed to identify Pulvis ellis urs, Capillary Wormwood Herbin, Atractylodis Macrocephalae Rhizoma and Glycyrrhizae Radix, and HPLC was used to determine the content of sodium tauroursodeoxycholic acid in Fufang Xiongdan Yinchen Granule. Kromail 100-5 C18 column (4.6 mm× 250 mm, 5 μm) in an oven at 25 ℃ was need, with a mobile phase consisting of methanol-0.03 mol/L sodium dihydrogen phosphate solution (60∶40, pH=4.4) and UV detector at 210 nm, the flow rate was 1.0 mL/min. Results Pulvis ellis urs, Capillary Wormwood Herbin, Atractylodis Macrocephalae Rhizoma and Glycyrrhizae Radix can be detected by TLC. The linearity of sodium tauroursodeoxycholic acid was obtained in the range of 0.182 1-1.821 mg (r=0.999 98). The average recovery was 100.39%, and RSD was 1.70% (n=6). Conclusion The method is simple, accurate, specific, and can control the quality of Fufang Xiongdan Yinchen Granule effectively.

RESUMO

AIMTo study whether Mel has protective role in liver c old preservation and its mechanism. METHODSTo study the role of Mel at the different concentration and time in liver cold preservation, we colle cted liver washing liquid and liver tissue from male SD rats. We analysed ALT, AST, LDH, HA, ET 1, MDA and SOD to see the function of liver cell and sinusoida l lining cell and mechanism of cell injury. At the same time, we studied the eff ects of Mel at different concentration and different time on liver cold preserva tion. RESULTS① The damage to liver function and change of its histology was aggravated with the prolongation of UW co ld preservation time, and injury of the sinusoidal lining cell was also aggravated gr adually. ② Indexes of the liver function and the sinusoidal lining cell functio n in the Mel solution cold preservation group were lower than that in the UW sol ution cold preservation group. These suggest protective role of Mel against live r injury caused by cold preservation. Its mechanism may be related to powerful a nti-oxidative effect of Mel. ③ The protective effect of Mel at 1?10 -5 m ol?L -1 ,1?10 -7 mol?L -1 was better than that of Mel at 1?10 -9 mol?L -1 . We found no obvious difference between the protection of Mel 1?10 -5 mol?L -1 and 1?10 -7 mol?L -1 indicati ng a concentration-effect relationship. CONCLUSIONProtective ef fect of UW solution used in clinic weakens gradually as cold preservation time increases. Mel solution has protective effect on liver cold preservation injury and is better than the simple UW solution. Its mechanism is related to anti-oxi dative effect of Mel.

Aim To provide experimental basis to study Human Cytomegalovirus(HCMV)infected disease and filtrate anti-virus drugs, HCMV acute interstitial pneumonia murine model was established. Methods Balb/c mice were infected with HCMV AD_ 169 strain. The rate of breath and weight of mice were observed, specific anti-HCMV antibody in serum was detected by ELISA, and viral isolation, Polymerase chain reaction(PCR) and pathological examination of lung tissue were performed aswell. RESUMO

AIM To investigate the effect of paeonol on proliferation and apoptosis of K 562. METHODS The inhibitory effect of paeonol on K 562 cells proliferation was assayed by MTT dye reduction. HE staining were used to observe the morphology of apoptotic cells, and Flow cytometric analysis were also used to analyze the K 562 apoptosis. RESULTS Pae at the concentration of 7.81~250 mg?L -1 significantly inhibited proliferation of K 562 cells in dose-dependent manner. Typical apoptotic changes were observed in K 562 cells under the light microscope. By FCM methods, the apoptotic rates of K 562 cells were increased from 10.01% to 34.16% after treatment of Pae at concentrations from 7.81 to 125 mg?L -1 for 24 h, which showed obvious concentration-effect relationship. The apoptotic rates of K 562 cells were also increased when Pae (6.25, 25 and 100 mg?L -1) was treated for 6, 12 and 24 h, which showed obvious time-effect relationship. CONCLUSION Paeonol may inhibit K 562 cell proliferation and induce its apoptosis.
